After incubation for 24h
1. Dilute and spread plate
2. Streak plate
3. Measure OD
4. Make 2 1ml pelletes each
5. Culture in new media

Serial dilution and spread plate
Used 2ml Eppendorf tubes, 10ul of culture to 990ul of peptone water. (10-2 dilution)
10ul of 10-2 dilution to 990ul of peptone water in new tube. (10-4)
Make 10-6 as well
Spread plate 100ul of 10-4. (LB)
Spread plate 10-6 in duplicates. (LB)

Streaked original culture onto LB plates.

Inoculated 0.5ml of overnight culture into new media.

Do plasmid loss test tomorrow. Autoclave toothpicks! And make agar asap, or as soon as the autoclave machine is available.

Ran gel on yesterday's RE digested plasmids.
Results: a 6+kb DNA for plasmid with 1 cut
a 4+kb DNA and a 1+kb DNA for plasmid with 2 cuts
The plasmid is pkd46. Yay!!
