Made LB broth, agar and EMB agar.
EMB agar is used to test for E.coli.
EMB agar 37.5g/l

Poured agar. Pour the smallest bottle of agar first, ie EMB agar, cos it cools the fastest.

Plasmid lost test
1. Draw 25 squares on petridishes (1 LB, 1 LB + amp for each strain)
2. Pick 1 colony with a toothpick, streak on LB + amp, and then streak on LB with the same toothpick. (replica plating)
3. Finish for all 25 squares
4. Incubate overnight
