Prepared agar and broth

1. Follow all instructions.
2. Make sure autoclave machine is available
3. Wash containers
4. Be careful when transferring powder into containers, don't spill onto weighing balance. Error ~0.02g
25g/l LB broth
15g/l agar
0.1% peptone water
50ug/ml ampicillin, stock: 100mg/ml, so add 500ul stock/l of broth
remember dilution: c1v1=c2v2
5. Add milliq water halfway, shake to make sure powder don't stick to container. Wait for foam to disappear before adding more water to the mark
6. Stick autoclave tape.
7. Autoclave. Make sure machine has water, and drain has the right amt of water. Loosen lid. Start autoclaving.
8. Wait for everything to cool
9. Take things out, keep broth in cold room
10. Take out petri dishes. Label.
11. Pour agar into petri dishes, pour enough to cover base. Shake petri a bit to make sure agar covers base. Ensure that agar doesn't touch lid
12. Leave for agar to solidify with lid of petri slightly open to allow evaporation
13. Close lid when agar is solidified, pack dishes into bag
14. Seal bag with tape, keep in cold room

Labelling: Write down name of stuff, date, my name, H/P

Safety briefing:
Found out where emergency showers are and how to use them.
Got locked into cold room.. Cold..
Practise opening doors without using hands.
Don't be too hardworking and work on weekends or after 8pm on weekdays.
Biopolis bus goes NUH or Holland.
There are security cameras everywhere.

Pipette tip don't touch anything wrong, like hands or bottle mouths.

Inoculated bacteria into shake flasks.
1. Use electronic pipette to pipette 50ml of broth into flask.
2. Add antibiotic from fridge into right broth.
3. Use clean pipette touch a typical distinct colony, touch broth in flask, swirl a bit. Throw pipette tip away.
4. Put flasks in that swirler thing with lids loose, 25 rpm or sth like that. 37 degrees overnight.
5. Clean up.
