Acetic acid assay
UV method (wavelength: 340nm)
Some enzymatic reaction that converts acetate to acetyl-CoA which combines with oxaloacetate (recall: Krebs cycle) to form citrate. Oxaloacetate and NADH is formed from reaction between L-malate and NAD+. This assay measures the amount of NADH formed by measuring absorbance at 340nm.
Run IV trial test
1. To samples, add 10ul of 0.02M NaOH, to create a slightly basic environment so that acetic acid exists as acetate ions which are more stable, less volatile.
2. Heat at 80degrees for 5 min to denature enzymes that might degrade acetate/ acetic acid
3. Centrifuge for 1min to remove any remaining cells inside

Follow instructions
1. Dissolve contents of bottle 2 (ATP, CoA, NAD) in 7ml H2O
2. Dissolve 1 bottle 4 in 0.25ml H2O
3. Dilute samples: concentration of acetic acid must be between 0.03g/l and 0.15g/l
4. Follow instructions, pipette right amount of all solutions and samples into 5ml cuvettes.
5. Read A0, A1.
6. Read A2 when reading stops changing, ie reaction has stopped
7. Calculate using formula given
