Microarray troubleshooting continued
1. Washed slides in 3 buffers, 5 min each
2. Dried by spinning for 2 min at 500g
3. Scanned
Strange results:
Very strong green spots for Arabidopsis? control genes on slides 1 and 2. No red on those spots. Strange because the cDNA was supposed to be labelled with Cye 5 which is a red dye. So we should see red instead of green. Furthermore when we scanned the gel yesterday with 625nm, we saw something. So there shouldn't be the problem of mislabelling of DNA.
Follow up:
Run gel and scan with Cye 3 frequency. Check if DNA is mislabelled.
Plasmid loss test that I've been doing for the past 2 weeks:
Got results until Day 7
0 loss until Day 3, then Day 7, about 20% loss for all (cells in LB and cells in LB+ampicillin). So exciting to see loss.
Did a negative control yesterday by streaking DH5a onto one of the LB amp plates. Nothing grew. Good, my ampicillin is still working.
Strange: Plasmid loss same extent and at the same rate in both LB and LB + amp, ampicillin is unnecessary in subculturing?
