Plasmid miniprep on Run VI samples. Measured plasmid concentration with Gene Quant.

Results of plasmid loss test:
No loss for time 5 and 5.5, Big M at time 11 can grow on LB amp, E.coli at time 11 can grow on LB amp

RNA extraction was done yesterday.
RNA purification today.
Follow modified protocol. Spin, add stuff, blah. Dissolved RNA in 60ul of water to make 10ug/ul. Final conc was about 6+ug/ul
Prepared stock of dNTP nd R9 (random nanomers)
Make a mastermix of buffer, dNTP, DTT and Superscript enzymes and 2ul of Ribolock to enable good mixing, should make a bit extra.
Add to RNA. Mix well. Add Cy3 and Cy5 to the right mixtures.
2 replicates and a Dye Swap
Incubation for 2h for reverse transcription

PCR MiniElute, elute in 15ul H2O, vacuum dry and dissolve in buffer..
